South Sudan Oil & Power 2026 to Bring Investors Directly to Juba’s Next Energy Opportunities 2026-10-25 · APO Group · Finance, Energy, Government & policy, Chemicals & materials, Mining & metals, Oil & gas https://energycapitalandpower.africa-newsroom.com/press/south-sudan-oil-and-power-2026-to-bring-investors-directly-to-jubas-next-energy-opportunities?lang=en For exploration companies, the opportunity starts with acreage. South Sudan’s Ministry of Petroleum currently lists 14 open blocks, including A1, A2, A3, A4, A5, A6, B1, B4, C1, C2, D1, D2, E1 and E2, while existing exploration acreage includes B3, 5B and B2. Investors can access geological and block information through the ministry’s data-viewing and acquisition process. The producing side of the market offers a different set of opportunities. South Sudan is targeting higher output from fields that have already been developed, creating demand for enhanced oil recovery, well intervention, drilling, production optimization, equipment and digital technologies. Greater Pioneer Operating Company (GPOC), for example, increased production to more than 60,000 barrels per day in May 2026, while national output was reported at around 174,000 bpd in June. The government has set longer-term ambitions of reaching 230,000 bpd in the short term and 450,000 bpd over time. That creates a market for oilfield service companies and technology providers able to work across mature assets, from seismic and subsurface services to drilling, artificial lift, production chemicals, maintenance and field automation. Nilepet’s own mandate covers exploration, development, operations, production and facilities across the country’s joint operating companies, underscoring the breadth of technical engagement available to prospective partners. Infrastructure is another area where commercial discussions are already moving. South Sudan’s experience with disruptions to crude exports through Sudan has reinforced the importance of resilient transportation and midstream systems. The United Nations documented the 2024 rupture and subsequent disruption to the Bashayer pipeline carrying Dar Blend crude. At the same time, the Ministry of Petroleum is seeking investment in pipelines, storage, distribution and related infrastructure. Downstream development adds another layer. South Sudan is pursuing additional refining capacity, including projects around Tharjiath, Thiangrial and other locations, with the stated objective of increasing domestic fuel supply and reducing reliance on imports. A 2025 government announcement also said a mini-refinery in Unity State had become operational, illustrating the movement from project proposals toward physical downstream assets. SSOP provides a setting for companies to meet directly with potential partners and decision-makers. The 2023 edition attracted more than 600 delegates from 24 countries and produced five strategic agreements covering areas including upstream exploration, drilling capacity, mining and regional energy cooperation. Participants included government ministers, international companies, financiers and energy institutions from Africa and beyond. For companies looking at South Sudan, that network matters. SSOP 2026 will put prospective investors and suppliers in the same room as the Ministry of Petroleum, Nilepet, operating companies, financiers, project developers, technology providers and regional energy stakeholders. In a market where access to decision-makers, local partners and project information can be as important as capital itself, two days in Juba can provide a concentrated opportunity to identify where commercial discussions can move next. Darolutamide is currently not approved for this treatment setting. These data will be presented at Presidential Symposium I at the ESMO Congress 2026 (LBA4, Saturday 24 October, 16:30 CET). At a median follow-up of 3.8 years, the interim analysis demonstrated a 58% reduction in the risk of metastasis or death with darolutamide plus ADT and RT compared with placebo plus ADT and RT (hazard ratio [HR] 0.42; 95% CI: 0.27–0.66; p=0.0001). The 4-year metastasis-free survival rate was 95% for patients receiving darolutamide compared with 88% for those receiving placebo. “Prostate cancer patients at a high risk of relapse face a considerable risk of recurrence and metastatic spread, making early, highly effective intervention vital,” said Dr. Tamim Niazi, Associate Professor of Oncology at McGill University, Radiation Oncologist at the Jewish General Hospital in Montreal, Canada. and Co-Lead Investigator of the study. “The clear improvement in metastasis-free survival seen in interim results from the DASL-HiCaP trial highlights the potential of adding darolutamide to standard androgen deprivation therapy and radiation therapy to help prevent or delay disease progression in patients who are at high risk of progression despite receiving primary curative therapy.” Secondary endpoints also showed consistent positive trends with darolutamide, with a 53% reduction in the risk of prostate-specific antigen progression free survival (PSA-PFS; HR 0.47; 95% CI: 0.33–0.67), and a 4-year PSA-PFS rate of 91% versus 83% with placebo. Prostate cancer-specific survival (PCSS) and overall survival (OS) hazard ratios were 0.58 (95% CI: 0.17–1.97; 99% vs 99% at 4 years) and 0.88 (95% CI: 0.49–1.59; 97% vs 95% at 4 years), respectively, with follow-up ongoing. The safety profile of darolutamide was consistent with its known tolerability, with adverse events being similar to the placebo group. Proportions of patients experiencing Grade ≥2 adverse events of special interest for darolutamide versus placebo were: fatigue/asthenia (14% vs 10%), hypertension (7% vs 9%), fractures (2% vs 3%), and cardiac events (3% vs 3%). Any Grade ≥3 adverse event occurred in 43% of patients in the darolutamide group versus 36% in the placebo group. "These new data highlight the potential of darolutamide to make a meaningful difference for patients earlier in the course of their prostate cancer,” said Christine Roth, Executive Vice President, Global Product Strategy and Commercialization at Bayer’s Pharmaceuticals Division. “By advancing darolutamide into earlier disease settings, we aim to reach more patients across the prostate cancer care continuum. We are proud to support ANZUP, the Sydney Clinical Trials Centre, CCTG and international partners in research as we work together with the potential to improve outcomes for men living with prostate cancer.” Follow-up of participants is continuing to provide further information on longer-term outcomes, including overall survival. About the DASL-HiCaP trial (ANZUP1801)  The DASL-HiCaP trial is an investigator-initiated, randomized, placebo-controlled, Phase III trial (NCT04136353) evaluating darolutamide added to standard-of-care ADT and locoregional radiation therapy in prostate cancer patients at high risk of recurrence. DASL-HiCaP was conducted in collaboration with the University of Sydney and ANZUP Clinical Trials Group and is the fourth Phase III trial for darolutamide. Following ARAMIS, ARASENS, and ARANOTE, which demonstrated darolutamide’s efficacy and safety in advanced prostate cancer, DASL-HiCaP represents the first Phase III study of darolutamide in early prostate cancer. A total of 1,106 participants across Australia, New Zealand, Canada, the United States, Ireland and the United Kingdom were randomized 1:1 to receive darolutamide (600 mg twice daily) or matching placebo for 96 weeks, combined with an LHRH analog for 96 weeks and RT to the prostate/prostate bed and pelvic lymph nodes. The primary endpoint was metastasis-free survival based on conventional imaging (MFS-CI). Secondary endpoints included prostate-specific antigen progression free survival (PSA-PFS), prostate cancer-specific survival (PCSS), and overall survival (OS). The protocol specified an interim analysis after 87 MFS-CI events; follow-up is ongoing to evaluate long-term overall survival. About the Sydney Clinical Trials Centre The Sydney Clinical Trials Centre is a flagship research centre of the Faculty of Medicine and Health at the University of Sydney that designs and manages clinical trials. This includes responsibility for study coordination, monitoring, data acquisition and management and statistical analysis. About Prostate Cancer Prostate cancer is the second most common cancer and the fourth most common cause of cancer death in men worldwide. Over 1.5 million men are diagnosed with prostate cancer each year and almost 420,000 die from the disease worldwide. By 2040, prostate cancer diagnoses are projected to increase to 2.9 million. Standard therapy for prostate cancer patients at high risk of recurrence combines radiation therapy with androgen deprivation therapy (ADT). However, despite this approach, a significant proportion of patients experience treatment failure, local recurrence, or distant metastases. Intensifying therapy early with an advanced androgen receptor inhibitor aims to improve long-term disease control and delay metastatic progression. About darolutamide (Nubeqa™) Darolutamide is an oral ARi with a distinct chemical structure that binds with high affinity to the androgen receptor and exhibits a strong antagonistic effect against the androgen receptor inhibiting the receptor function and growth of prostate cancer cells. Additionally, preclinical models and neuroimaging data in healthy humans support darolutamide's low potential for blood-brain barrier penetration and low potential for clinically relevant drug-drug interactions. Darolutamide, under the brand name Nubeqa™ plus androgen deprivation therapy (ADT), is already approved in more than 85 markets around the world, including the US, EU and China. It is approved for the treatment of adult patients with mHSPC without docetaxel in 75 markets and with docetaxel in more than 85 countries, and for adult patients with nmCRPC at high risk of developing metastatic disease in more than 85 countries. Darolutamide is developed jointly by Bayer and Orion Corporation, a globally operating Finnish pharmaceutical company. Darolutamide demonstrated efficacy and tolerability in pivotal Phase 3 studies, supporting its indications in nmCRPC and mHSPC with and without chemotherapy, with a favorable safety profile that offers an advancement in treatment management for physicians and patients. A robust clinical development program is underway investigating darolutamide across various stages of prostate cancer. Current ongoing trials include the Phase III ARASTEP trial evaluating the efficacy of darolutamide plus ADT compared to ADT alone in hormone-sensitive prostate cancer (HSPC), in patients with high-risk biochemical recurrence (BCR) who have no evidence of metastatic disease by conventional imaging and a positive PSMA PET/CT at baseline. As these developments open new opportunities for regional petroleum trade, the company's CEO and Managing Director David Bird has joined the South Sudan Oil & Power (SSOP) 2026 conference as a speaker. Taking place in Juba on November 24-25, SSOP returns for its seventh edition as the nation’s premier platform for investment, dealmaking and policy. Bird’s participation comes as South Sudan looks to translate its resource potential into renewed exploration, higher production and greater participation in regional petroleum value chains. With major downstream investment reshaping East Africa's energy market, SSOP 2026 offers an opportunity for producers, refiners and investors to establish the partnerships needed to advance regional energy trade and unlock new commercial opportunities. The event arrives at a strategic time for Dangote. On September 30, the company officially broke ground on a 700,000-barrel-per-day (bpd) refinery in Lamu, Kenya. Set to come online in 2029, the facility is designed to supply gasoline, diesel and jet fuel to East African markets, with regional governments offered a combined 30% stake in the project. The project aims to replicate Dangote’s 650,000-bpd Nigerian facility, which is currently operating at full capacity and is set to expand to 1.2 million bpd. A delegation from South Sudan attended the Lamu groundbreaking ceremony, underscoring the country’s commitment to strengthening regional petroleum ties. https://apo-opa.co/4zQaHcU Beyond the Lamu facility, Dangote is strengthening its footprint across East Africa’s distribution networks. Just this month, the company acquired a 25% stake in KPC, securing access to established infrastructure transporting petroleum products from Kenya's coast to regional markets, including Uganda, Rwanda, South Sudan and the Democratic Republic of the Congo. Following the $335 million transaction, Dangote plans to invest an additional $1.5 billion to expand KPC’s approximately 1,300-km pipeline network, bringing its total planned investment to $1.8 billion. https://apo-opa.co/4zS8fCN For South Sudan, East Africa's only established commercial crude oil producer, Dangote's expansion introduces new opportunities for regional petroleum integration. The country holds approximately 3.5 billion barrels of proven oil reserves and is seeking to increase production through new exploration, redevelopment of mature fields and investment in supporting infrastructure. While no crude supply agreement between South Sudan and Dangote has been announced, the development of additional regional refining capacity could create opportunities for future feedstock supply arrangements, subject to commercial viability, infrastructure and crude compatibility. Bird brings extensive experience across refining, petrochemicals and integrated energy infrastructure to SSOP 2026. Before joining Dangote in 2025, he served as CEO of OQ8, the $9.5 billion Duqm refinery joint venture in Oman, overseeing the development and commissioning of major refining, pipeline and terminal infrastructure. He also held senior leadership positions at Shell, spanning upstream production, downstream operations and global energy supply chains. His participation at SSOP 2026 will bring expertise in large-scale refining investment, project delivery and regional petroleum market development to discussions on South Sudan's next phase of growth. https://apo-opa.co/3WotMnT As the country's official energy event, held in partnership with the Ministry of Petroleum, SSOP 2026 provides a platform for advancing discussions around regional trade, refining and infrastructure investment. The conference brings together international and national oil companies, refiners, trading companies, infrastructure developers, financiers and technology providers to negotiate partnerships, mobilize investment and connect South Sudan's petroleum resources with regional and international markets. Day One of MSGBC Oil, Gas & Power 2026 will bring these priorities together, with a program focused on investment, project development, regional cooperation and the commercial opportunities emerging across the MSGBC basin. Taking place December 1-3 at the Centre International de Conférences Abdou Diouf (CICAD) in Dakar, Senegal, the fifth edition of the conference and exhibition will connect policymakers, energy companies, investors, financiers and technical partners from across Mauritania, Senegal, The Gambia, Guinea-Bissau and Guinea-Conakry. Day One of the event – taking place on December 2 – will open with a Presidential and Ministerial programme led by H.E. Bassirou Diomaye Faye, President of Senegal. Senior representatives from the African Energy Chamber, Gas Exporting Countries Forum, African Petroleum Producers’ Organization, bp and Woodside will join the opening, setting the policy and investment context for discussions on the basin’s growing role in global energy markets. A Ministerial Dialogue will then bring together energy leaders from Senegal, Mauritania, The Gambia, Guinea-Bissau and Guinea-Conakry to examine how the region can accelerate exploration and production while developing the infrastructure needed to monetize its natural gas resources. Regional integration, cross-border cooperation, innovation and storage will form part of the discussion as governments work to translate resource development into energy security and economic growth. The investment agenda will continue with the CEO Regional Leadership Panel, “Is MSGBC the Next Global Gas Hotspot?” Executives from companies including bp, Chevron, Energean, Woodside Energy and Africa Finance Corporation will consider what is driving investment into the region’s gas sector and how MSGBC markets can compete for international capital. The program will then turn to the basin’s resource potential and project pipeline. “The Future of the MSGBC Basin: Where the Next Billion Barrels and Trillions of Cubic Feet Will Come From” will examine emerging exploration opportunities, followed by “From Discovery to Development: Projects Moving Toward FID,” highlighting projects approaching final investment decisions and the milestones needed to unlock financing. Natural gas will take center stage in “Gas as the Engine of Regional Transformation,” with discussions covering gas-powered electrification, regional gas markets, transmission infrastructure and the role of gas in supporting fertilizer, petrochemical and industrial development. The commercial focus will continue through strategic dealmaking and signing ceremonies, where announcements and agreements will cover upstream partnerships, infrastructure investments, financing arrangements and energy cooperation frameworks. The day will culminate in the MSGBC Oil, Gas & Power Gala Dinner & Awards Ceremony, bringing together government officials, investors and industry leaders for an evening of networking and recognition. Following a full day of policy discussions, investment dialogue and commercial activity, the Gala will provide a further opportunity to build relationships with the companies and institutions shaping the region’s energy future. With major projects now requiring capital, infrastructure and strategic partnerships to move into their next stages, Day One will showcase the MSGBC region as an increasingly important destination for energy investment and a market where resource development is being linked directly to regional economic growth. MSGBC Oil, Gas & Power 2026 takes place December 1-3 at the CICAD in Dakar, Senegal, under the High Patronage of President Bassirou Diomaye Faye. The recognized companies - Procter & Gamble (P&G), Nestlé, Mars, AstraZeneca, ITC, and Bekaert - were celebrated for driving decision velocity, co-innovation, and impactful business results. Supply Chain Innovation & Excellence Award: Nestlé and Bekaert Nestlé, one of the world's largest food and beverage companies, and Bekaert, a global market and technology leader in material science of steel wire transformation and coating technologies, received OMP's Supply Chain Innovation & Excellence award for their breadth of deployment and depth of co-innovation. Nestlé aligns its supply network at the strategic, tactical, and operational levels on OMP's Unison Planning™ platform, connecting forecasting, planning, and replenishment end-to-end. As more routine work becomes touchless, planners can focus on the decisions that drive the business, and Nestlé continues to roll out best practices throughout the organization. Bekaert integrated scenario planning directly into its S&OP, shifting teams from reactive firefighting to proactive, value-driven conversations. In its green hydrogen business, what-if modeling protects mid- and long-term strategic control. Furthermore, planners use Unison Companion, OMP's generative AI assistant, to generate tables, compare scenarios, and retrieve information in natural language. "This award recognizes the ongoing transformation of our end-to-end supply chain planning," says Bo Yang, Senior Manager, Global Supply Chain Process Management at Bekaert Rubber Reinforcement. "We've moved from manual Excel calculations and individual experience to standardized planning on OMP's Unison Planning™ platform. Our decisions are now backed by data, and our planning accuracy has improved significantly." Long-Term Partnership Award: P&G and AstraZeneca P&G, with operations in approximately 70 countries, joins global biopharma leader AstraZeneca in accepting the award for proving that value compounds the longer the partnership runs. P&G celebrates 27 years of partnership with OMP dating back to 1999, and operates a planning network spanning more than 100 manufacturing sites and 200 distribution centers. The company has consolidated overlapping planning processes, with the majority of planning now handled touchlessly. "Receiving OMP's Long-Term Partnership Award reflects years of trust and teamwork between our teams," says Jen Aberilla, Director, Regional Supply Network Operations Planning at P&G. "We're proud of what we've achieved together and look forward to building on it." AstraZeneca has been working with OMP since 2021, replacing siloed, spreadsheet-based planning with a single capacity-constrained network plan. Most recently, the company has been collaborating with OMP on AI‑driven planning and advanced scenario modeling, allowing planners to focus on scenarios and exceptions. This has resulted in improved supply reliability and service levels, inventory reduction, and productivity savings. Community Engagement Award: Mars and ITC ITC's Paperboards & Specialty Papers Division, part of the Indian multi-business enterprise ITC Limited, and Mars, a global giant in confectionery, food products, and pet care services, received the Community Engagement Award for their active role in OMP Community events and councils. ITC was recognized for its commitment to openly sharing its planning transformation with the wider supply chain community. By presenting its experience to industry peers, ITC has given them a candid look at how its journey unfolds in practice, with lessons other planning teams can apply. Mars is a committed participant in the OMP Community councils and in governance through the executive board. Working with OMP, Mars has lowered inventory, consolidated its warehouse network, improved factory equipment effectiveness, and increased forecast accuracy. "The world's most demanding manufacturers plan with OMP, and it is a privilege to see those relationships deepen over 10, 20, and 27 years. The results they achieve through this community are concrete: inventory goes down, forecast accuracy goes up, and decisions happen faster. Congratulations to each of them!" says Philip Vervloesem, Chief Commercial and Markets Officer at OMP. About OMP OMP helps companies facing complex planning challenges to excel, grow and thrive by offering the best digitized supply chain planning solution on the market. In total, around fifty Cameroonian service members refined their capabilities in countering unconventional threats and explosive hazards. From September 28 to October 2, 2026, twenty personnel completed a specialized course on Nuclear, Radiological, Biological, and Chemical (CBRN) hazards. Led by French Army instructors, the session provided solid tactical and technical foundations, enhanced risk identification, and refined crisis management procedures. Particular emphasis was placed on mitigating risks associated with chemical improvised explosive devices. This foundational training paves the way for sustained cooperation, which remains vital for knowledge sharing and operational effectiveness. Building on this momentum, thirty bomb disposal experts completed an advanced Explosive Ordnance Disposal (EOD) course from September 23 to October 16, 2026. To meet the strategic need for highly skilled explosive ordnance disposal technicians, the Military Engineering Corps organized this month-long program in coordination with the Cameroonian and French armed forces. Co-conducted by two Cameroonian and three French instructors, the curriculum progressed from advanced theory and practical drills with specialized equipment to field exercises using live explosives. This dual training effort highlights the leadership’s commitment to maintaining peak operational readiness, ensuring public safety, and tackling complex security threats in the field. TAIPEI, TAIWAN, Octob ———————————————————————————————————————————————————————————— Roche presents new pivotal trial data across broad neuroimmunology portfolio at MSToronto2026 2026-10-21 · Roche · Healthcare, Automotive, Travel, Biotech & pharma, Sustainability, Government & policy, Medical devices, Chemicals & materials https://www.roche.com/media/releases/med-cor-2026-10-21 Fenebrutinib FENtrepid and FENhance 1 & 2 late-breaking data demonstrate a therapeutic effect on chronic active lesions, addressing a primary driver of long-term disability progression in multiple sclerosis (MS) Fenebrutinib FENhance 1 & 2 Phase III pooled data show a 55% reduction in annualised relapse rate (ARR) vs active comparator Following European Commission paediatric approval on 15 October, new Ocrevus OPERETTA 2 Phase III data show 90% of children aged 10–17 remained relapse-free over two years Featured in "Best of MSToronto2026", Enspryng METEOROID Phase III data highlight significant attack reduction and improved functional recovery in MOGAD, an underserved autoimmune disease Basel, 21 October 2026 - Roche (SIX: RO, ROP; OTCQX: RHHBY) announced today new Phase III data for investigational fenebrutinib in multiple sclerosis (MS), demonstrating reduced markers of chronic brain inflammation as well as more comprehensive disease control versus active comparators.1-3 These findings, alongside new Ocrevus® (ocrelizumab) and Enspryng® (satralizumab) data, will be featured across more than 40 abstracts at MSToronto2026 (10th ACTRIMS- ECTRIMS Meeting), held 21 to 23 October in Toronto. Highlights include a fenebrutinib late-breaking presentation and an Enspryng presentation in the “Best of MSToronto2026.4 “Addressing long-term disability progression to preserve people’s independence remains a critical challenge in neuroinflammatory diseases, and particularly in multiple sclerosis,” said Levi Garraway, M.D., Ph.D., Roche’s Chief Medical Officer and Head of Global Product Development. “Our recent data indicate that fenebrutinib can inhibit disease activity inside and outside the central nervous system, underscoring its potential to transform care for people living with relapsing and primary progressive forms of multiple sclerosis.” Fenebrutinib targets progressive biology inside the brain New data from the pivotal trials in RMS (FENhance 1 and 2) and PPMS (FENtrepid) support the potential of fenebrutinib as the first and only high-efficacy oral medicine for both forms of MS. Reduction of chronic active lesions (Late-breaking oral #O137): In the first prospective demonstration of a therapeutic effect on chronic active lesions in MS Phase III clinical trials, data show fenebrutinib reduced the volume of slowly expanding lesions (SELs) by up to 30% (p<0.001) in RMS (over 96 weeks) and PPMS (over 120 weeks), along with a 62% (p=0.001) reduction in accumulation of paramagnetic rim lesions (PRLs) in RMS, versus active comparators. These findings demonstrate that fenebrutinib directly targets chronic neuroinflammation in the brain, a primary driver of disability progression in MS.1 Comprehensive disease control (Poster #P1434): In the pooled RMS trials, three quarters (75%) of fenebrutinib-treated patients showed no evidence of disease activity (NEDA-3) meaning zero relapses, zero MRI activity and zero worsening disability at nearly two years, a 70% reduction (p<0.001) versus the comparator. These results show fenebrutinib’s potential to suppress disease activity and delay progression.2 Preserving brain volume and function (Oral #O089): Pooled RMS data show fenebrutinib reduced the annualised relapse rate (ARR) by 55% versus the comparator (p<0.001) over 96 weeks. In addition, fenebrutinib reduced annual total brain volume loss by 23% (p<0.001) and 12-week composite confirmed disability progression by 16% (p=0.04). These findings demonstrate fenebrutinib’s potential to control relapses, to slow brain tissue loss caused by neuronal cell death and to preserve function.3 Ocrevus provides consistent benefits across all age groups New data show that Ocrevus provides significant and consistent efficacy across a broad MS patient population, from children 10 years of age to older adults. Two-year data and immune function in paediatric MS (3 posters on the Phase III OPERETTA 2 study): 90% of children with relapsing-remitting MS (RRMS) treated with Ocrevus, who were 10-17 years old at trial entry, remained free of relapses over two years (#P1439).5 Ocrevus selectively depleted target B cells with little to no effect on other immune cells (#P1407), while preserving pre-existing immunity to common childhood vaccines in 81–100% of patients (#P1415). The European Commission approved Ocrevus in this indication on 15 October, following FDA approval in May.6,7 Two-year data in patients >55 years old (Poster #P1426 on the Phase IIIb CONSONANCE study): 53% of participants over 55 years old treated with Ocrevus showed no disability progression after two years, similarly to younger participants (55%), showing Ocrevus maintains its effect on progression as people age.8 Switching from other treatments to Ocrevus: Real-world data show progression was the leading reason for patients switching from ofatumumab to Ocrevus (ePoster #EP2733).9 A separate analysis of patients treated with Ocrevus or ofatumumab for one year show Ocrevus was associated with lower MRI lesion activity (Poster #P0305).10 Enspryng reduces risk of attack and is associated with improved recovery Exploratory analyses of the pivotal METEOROID trial suggest that in addition to significantly lowering the risk of relapse (AAN 2026), Enspryng was associated with improved recovery and reduced disability progression, which is critical to prevent permanent damage to the optic nerves, spinal cord or brain in myelin oligodendrocyte glycoprotein antibody-associated disease (MOGAD). These findings reinforce Enspryng’s potential to become the first FDA-approved treatment for MOGAD, with a decision in the U.S. expected in January 2027.4,11 Relapse recovery (Oral #O035): Patients treated with Enspryng achieved numerically superior recovery at 12 weeks post relapse versus placebo (median change in Expanded Disability Status Scale [EDSS] of -2 vs -0.5). Overall function was stable (median 0) with Enspryng, compared to worsening (median 0.5) with placebo, as measured with the EDSS at the end of the double-blind period or first relapse.4  About fenebrutinib Fenebrutinib is an investigational oral, central nervous system (CNS)-penetrant, reversible and non-covalent Bruton’s tyrosine kinase (BTK) inhibitor with an optimised pharmacokinetics (PK) profile. Fenebrutinib can act throughout the body and also cross the blood-brain barrier into the CNS to target chronic inflammation. It is uniquely designed to target relapsing and progressive biology by inhibiting cells in the immune system known as B cells and microglia. Targeting B cells helps control the acute inflammation that causes relapses, while targeting microglia inside the brain addresses the chronic damage that is thought to drive long-term disability progression. While most current BTK inhibitors are covalent and irreversible, meaning they form a permanent chemical bond with the enzyme, fenebrutinib is non-covalent and reversible, meaning it binds and then eventually releases the enzyme. About Ocrevus® (ocrelizumab) Ocrevus is a humanised monoclonal antibody designed to target CD20-positive B cells, a specific type of immune cell thought to be a key contributor to myelin (nerve cell insulation and support) and axonal (nerve cell) damage. Ocrevus IV and Ocrevus subcutaneous (SC; marketed as Ocrevus Zunovo® [ocrelizumab hyaluronidase-ocsq] in the U.S.) are the only therapies approved for both RMS (including relapsing-remitting multiple sclerosis [RRMS] and active, secondary progressive multiple sclerosis [SPMS], as well as clinically isolated syndrome [CIS] in the U.S.) and primary progressive multiple sclerosis (PPMS). Both Ocrevus IV and SC are administered every six months. The initial IV dose is given as two 300 mg infusions two weeks apart with subsequent doses given as single 600 mg infusions. Ocrevus SC is given as a single 920 mg subcutaneous injection every six months. About Enspryng® (satralizumab) Enspryng, developed by Chugai, a member of the Roche Group, is a humanised monoclonal antibody that binds to both soluble and membrane-bound interleukin-6 receptors (IL-6R), thereby inhibiting IL-6 mediated signalling, a key pathway involved in inflammation. Enspryng incorporates Chugai’s proprietary antibody recycling technology, which enables pH-dependent binding to IL-6R and repeated engagement of the target, extending antibody circulation and supporting sustained inhibition of IL-6 mediated signalling. Enspryng is the first and only IL-6R inhibitor treatment currently approved in approximately 90 countries for neuromyelitis optica spectrum disorder (NMOSD), including in the U.S. and E.U., with a well-established safety profile in over 16,000 patients. Enspryng was also recently approved for the treatment of thyroid eye disease (TED) in the U.S. Roche is committed to developing Enspryng in additional neurological autoimmune and inflammatory diseases that may benefit from inhibition of IL-6 mediated signalling, including myelin oligodendrocyte glycoprotein antibody-associated disease (MOGAD) and autoimmune encephalitis (AIE). Enspryng can be administered by a person living with TED or a caregiver following training from a healthcare provider.1 “Today’s FDA approval of Enspryng provides an important new option for people with thyroid eye disease, who have long faced limited treatment choices,” said Levi Garraway, MD, PhD, Roche’s Chief Medical Officer and Head of Global Product Development. “As the first subcutaneous treatment for this condition, Enspryng offers a positive safety and efficacy profile and the potential for at-home administration.” "Treating thyroid eye disease requires balancing efficacy, safety, and the practical realities of long-term treatment,” said César A. Briceño, MD, Clinical Trial Investigator and Associate Professor of Ophthalmology at the Scheie Eye Institute, University of Pennsylvania. “As a clinical trial investigator, I have seen firsthand the potential of targeted therapies such as Enspryng. The availability of an effective subcutaneous treatment option could provide greater flexibility for both patients and physicians while reducing the logistical burden associated with infusion-based therapies. Enspryng has the potential to meaningfully expand our therapeutic options for patients with thyroid eye disease." The totality of data from the pivotal Phase III SatraGO programme (SatraGO-1 and SatraGO-2) provided clinically meaningful improvements in patients with TED.1,2 While SatraGO-1 did not meet its pre-specified primary efficacy endpoint at Week 24, SatraGO-2 achieved significant and clinically meaningful difference in proptosis response (52.9% in the Enspryng group compared to 23.4% in the placebo group) and in diplopia reduction (60.7% in the Enspryng group compared to 25.8% in the placebo group) in the active TED population.1,2 Consistent results were achieved in the overall population (including both active and chronic patients) in SatraGO-2.1,2 No new safety signals were identified in the SatraGO trials, with Enspryng’s safety profile consistent with its known profile in neuromyelitis optica spectrum disorder (NMOSD).1,2 Full results were shared at the American Society of Ophthalmic Plastic and Reconstructive Surgery (ASOPRS) Fall Scientific Symposium in October 2025.2 “Thyroid eye disease not only affects a person’s eyes through bulging and vision problems, but it also takes a profound emotional toll on confidence, identity, relationships and daily life,” said Christine Gustafson, Chief Executive Officer of the TED Community Organization. “Until now, our community has had very limited choices. The approval of Enspryng represents a critical turning point through a convenient, targeted option.” The approval in TED, following a priority review designation, marks the second indication for Enspryng, following an approval in neuromyelitis optica spectrum disorder (NMOSD) in 2020.1,8,9 This follows a recent acceptance by the FDA for priority review in myelin oligodendrocyte glycoprotein antibody-associated disease (MOGAD).10 Enspryng is also being studied in additional neurological autoimmune and inflammatory diseases that may benefit from inhibition of IL-6 mediated signalling. About SatraGO-1 and -2 SatraGO-1 (NCT05987423) and -2 (NCT06106828) are identically designed, Phase III, randomised, placebo-controlled, multicentre studies to determine the efficacy, durability and tolerability of Enspryng for the treatment of adults with active, moderate‑to‑severe TED and chronic TED. The studies enrolled a total of 258 patients from 19 countries. Participants were randomised 1:1 to receive Enspryng or placebo. The primary endpoints were the proportion of participants with active, moderate-to-severe TED who achieved an at least 2 mm reduction in proptosis in the study eye from baseline at week 24.2,3 About thyroid eye disease (TED) TED, also known as Graves’ ophthalmopathy, is a complex inflammatory autoimmune disease, affecting the area around the eyes and the eyes themselves, that can be sight-threatening, debilitating and disfiguring.4,5 The most common symptoms are redness, swelling of the eyes, eyelid retraction, appearance of a stare, bulging of one or both eyes (proptosis), double vision (diplopia) and pain.4,6 TED is a progressive rare disease that affects approximately 155 people out of every 100,000.3-5,7 It most commonly occurs in people with hyperthyroidism, approximately 50% of whom experience at least mild TED, but it can also affect people with hypothyroidism or normal thyroid function.4,11 Despite existing approved treatments for TED, a medical need remains for therapies that are effective, well-tolerated, and have a convenient route of administration.3 About Enspryng® (satralizumab) Enspryng, developed by Chugai, a member of the Roche Group, is a humanised monoclonal antibody that is designed to bind to both soluble and membrane-bound interleukin-6 receptors (IL-6R), thereby inhibiting IL-6 mediated signalling, a known key pathway involved in inflammation. Enspryng incorporates Chugai’s proprietary antibody recycling technology, which enables pH-dependent binding to IL-6R and repeated engagement of the target, extending antibody circulation and supporting sustained inhibition of IL-6 mediated signalling.1-3,12 Enspryng is the first and only IL-6R inhibitor treatment currently approved in approximately 90 countries for neuromyelitis optica spectrum disorder (NMOSD), including in the European Union and United States, with a well established safety profile in over 16,000 patients.1,13 Roche is committed to developing Enspryng in additional neurological autoimmune and inflammatory diseases that may benefit from inhibition of IL-6 mediated signalling, including autoimmune encephalitis (AIE) and myelin oligodendrocyte glycoprotein antibody-associated disease (MOGAD). The FDA granted priority review of the Enspryng supplemental Biologics License Application (sBLA) for MOGAD in September 2026 with an approval decision expected in January 2027.10 Enspryng has orphan drug designation in the United States and European Union for NMOSD and MOGAD and in the United States for anti-NMDA receptor autoimmune encephalitis (anti-NMDAR AIE) and leucine-rich glioma-inactivated 1 autoimmune encephalitis (LGI1 AIE). The filing acceptance is based on data from the comprehensive clinical programme, including the Phase III FENhance 1 and 2 RMS studies and the Phase III FENtrepid PPMS study.1,2 "People living with multiple sclerosis need treatments that go beyond controlling relapses to help preserve function and independence as the disease progresses," said Levi Garraway, M.D., Ph.D., Roche’s Chief Medical Officer and Head of Global Product Development. "Three Phase III studies have demonstrated the potential for fenebrutinib to address both relapsing and progressive disease, thereby bringing us closer to an oral treatment that could make a meaningful difference across the MS spectrum." “Ocrevus transformed how we treat MS, helping more than 525,000 people live a life less defined by their disease. Yet, over a third of all people with MS still receive lower-efficacy treatment,” said Teresa Graham, Roche’s Chief Executive Officer, Pharma. “If approved, fenebrutinib could open a new chapter as the first high-efficacy oral therapy for both relapsing and primary progressive MS, helping to control disease activity while giving people more flexibility and choice.” Concurrently addressing the life-disrupting relapses and long-term disease progression – the slow, steady loss of physical and mental abilities over time – remains one of the greatest challenges in MS.4 Fenebrutinib is designed to act throughout the body and to cross the blood-brain barrier into the central nervous system to target both the acute inflammation that causes relapses and the chronic inflammation that is thought to drive disability progression.1,2 The NDA for fenebrutinib is supported by three positive Phase III studies: The FENhance 1 and 2 RMS studies showed that fenebrutinib significantly reduced relapses and both active and chronic brain lesions compared with standard of care, teriflunomide. Fenebrutinib reduced the annualised relapse rate (ARR) by 51.1% (p<0.001) in FENhance 1 and 58.5% (p<0.00001) in FENhance 2 compared with teriflunomide over 96 weeks. This equates to patients having approximately one relapse every 17 years, which is the lowest relapse rate seen in Phase III MS studies. In addition, measures of disability progression, including 12-week composite confirmed disability progression (cCDP12), showed consistent positive trends favouring fenebrutinib over teriflunomide.1 The FENtrepid PPMS study showed fenebrutinib met its primary endpoint of non-inferiority compared with Ocrevus® (ocrelizumab), the current standard of care and only approved medicine for PPMS, in reducing disability progression. Fenebrutinib numerically reduced the risk of progression by 12% compared to Ocrevus, as measured by the time to onset of cCDP12 (hazard ratio [HR] 0.88; 95% confidence interval [CI]: 0.75, 1.03), with curves separating as early as 24 weeks. A consistent treatment effect on disability progression was observed across patient subgroups, including those without active inflammation, and for the entire treatment duration after 24 weeks.2 The overall rate of serious adverse events for fenebrutinib and teriflunomide, respectively, was 9% and 9% in FENhance 1 and 11% and 6% in FENhance 2. The overall rate of serious adverse events in FENtrepid was 19% for fenebrutinib and 19% for Ocrevus. Liver enzyme elevations were comparable between the fenebrutinib and teriflunomide arms in the RMS studies and observed more often with fenebrutinib compared to Ocrevus in the PPMS study. While an imbalance in reported fatalities was observed across the three pivotal studies, the deaths occurred at different timepoints and had various causes.1,2 Overall, fenebrutinib has shown a manageable safety profile across the three Phase III trials and earlier studies, with a large safety database including more than 2,700 study participants.1,2,5 About fenebrutinib Fenebrutinib is an investigational oral, central nervous system (CNS)-penetrant, reversible and non-covalent Bruton’s tyrosine kinase (BTK) inhibitor with an optimised pharmacokinetics (PK) profile. About multiple sclerosis Multiple sclerosis is a chronic disease that affects more than 3 million people worldwide. People with all forms of multiple sclerosis experience disease progression from the beginning of their disease. Therefore, an important goal of treating multiple sclerosis is to slow, stop and ideally prevent progression as early as possible. Approximately 85% of people with multiple sclerosis are initially diagnosed with relapsing-remitting multiple sclerosis (RRMS). Relapsing forms of the disease (RMS) include RRMS and active secondary progressive MS, and people with RMS experience relapses and worsening disability over time. Primary progressive multiple sclerosis (PPMS) is a debilitating form of the disease marked by steadily worsening symptoms but typically without distinct relapses or periods of remission. Approximately 15% of people with multiple sclerosis are diagnosed with the primary progressive form of the disease. Until the FDA approval of Ocrevus®, there had been no FDA-approved treatments for PPMS, and Ocrevus is still the only approved treatment for PPMS.
