FDA approves Roche’s Tecentriq in combination with a fluoropyrimidine and oxaliplatin for the adjuvant treatment of a certain type of stage III colon cancer

FDA approval is based on results from the landmark phase III ATOMIC study, published in The New England Journal of Medicine, which showed Tecentriq plus chemotherapy reduced the risk of disease recurrence or death by 50% compared to chemotherapy alone1 First approved adjuvant immunotherapy regimen for stage III dMMR colon cancer, positioning Tecentriq as a new post-surgery standard of care Nearly 25% of all colon cancers are diagnosed at stage III, and almost one in three patients with stage III colon cancer experience a recurrence within five years2,3 Approval marks the twelfth indication for Tecentriq in the US Basel, 9 October 2026 - Roche (SIX: RO, ROP; OTCQX: RHHBY) announced today that the US Food and Drug Administration (FDA) has approved Tecentriq® (atezolizumab) and Tecentriq Hybreza® (atezolizumab and hyaluronidase-tqjs) in combination with a fluoropyrimidine and oxaliplatin for the adjuvant treatment of stage III deficient DNA mismatch repair (dMMR) colon cancer, a type of tumour characterised by high mutation rates. This approval marks the twelfth indication for Tecentriq in the US. “This approval provides a new adjuvant treatment option for people with stage III dMMR colon cancer who face a high risk of disease recurrence,” said Levi Garraway, MD, PhD, Roche’s Chief Medical Officer and Head of Global Product Development. “Tecentriq combined with chemotherapy has the potential to become the new standard of care as the first approved immunotherapy-based regimen for this setting.” “Until now, the adjuvant standard of care for stage III colon cancer has not differed based on mismatch repair status,” said Frank A. Sinicrope, MD, Professor of Oncology at the Mayo Clinic and US Principal Investigator for the Alliance ATOMIC trial. “This milestone represents an important therapeutic advance that leverages immunotherapy to target the specific biology of dMMR stage III colon cancer after surgical resection. By reducing the risk of disease recurrence or death by 50%, atezolizumab plus chemotherapy is a practice-changing regimen that can improve clinical outcomes of this group of patients living with colon cancer, one of the most common cancers and a leading cause of cancer-related death.” The FDA approval is based on the landmark phase III ATOMIC study, recently published in The New England Journal of Medicine. ATOMIC demonstrated that adding Tecentriq to modified (m)FOLFOX6 chemotherapy reduced the risk of disease recurrence or death by 50%, compared to chemotherapy alone for people with stage III dMMR colon cancer, as determined by an immunohistochemistry test, such as the VENTANA® MMR RxDx Panel.1 The 36-month disease-free survival was 86% for Tecentriq combined with mFOLFOX6 compared with 76% in the mFOLFOX6 alone group. The safety profile was consistent with previous studies of Tecentriq and mFOLFOX6.1 Colon cancer remains one of the world's most common and deadliest tumours.4 Over one million people are diagnosed globally each year, and nearly 25% of all colon cancers are diagnosed at stage III.2-4 Despite surgery and chemotherapy, nearly 30% of stage III patients experience disease recurrence within five years.2,3 Approximately 15% of colon cancer patients present with dMMR or microsatellite instability-high (MSI-H) tumours, which indicate a higher mutation rate and thus have the potential to respond to immunotherapy.5 Roche is pursuing further regulatory filings for Tecentriq, including with the European Medicines Agency, to bring this immunotherapy-based adjuvant option to patients with dMMR colon cancer worldwide as soon as possible. About the ATOMIC study ATOMIC (A021502, NCT02912559) is a phase III, randomised, open-label, multicentre study investigating the addition of Tecentriq® (atezolizumab) to modified (m)FOLFOX6 chemotherapy (a combination of folinic acid, fluorouracil, and oxaliplatin) in patients with stage III colon cancer who have a deficiency in DNA mismatch repair (dMMR). The trial enrolled 712 patients. Participants were randomised 1:1 to receive either mFOLFOX6 plus Tecentriq for 12 cycles (six months) followed by Tecentriq monotherapy for 13 cycles (an additional six months), or mFOLFOX6 alone for 12 cycles. The primary endpoint is disease-free survival (DFS). The global study in the US was sponsored by the US National Cancer Institute (NCI), part of the National Institutes of Health, and led by the Alliance for Clinical Trials in Oncology (Alliance) with participation from other NCI-funded National Clinical Trials Network (NCTN) groups. Genentech, a member of the Roche Group, provided support for the study through a Cooperative Research and Development Agreement between NCI and Genentech. The trial was led globally by Frank A. Sinicrope, MD, Professor of Oncology at the Mayo Clinic and US Principal Investigator within the Alliance. German participation in this major international academic collaboration was enabled through close cooperation between the Alliance and the AIO-Studien-gGmbH, in partnership with the AIO (Arbeitsgemeinschaft Internistische Onkologie) of the German Cancer Society (Deutsche Krebsgesellschaft, DKG), under the national leadership of Prof. Dr. Anke Reinacher-Schick (Ruhr-Universität Bochum). AIO-affiliated study sites enrolled patients during the later recruitment phase, ensuring access for eligible patients in Germany. About Tecentriq® (atezolizumab) Tecentriq is a monoclonal antibody designed to bind with a protein called PD-L1, which is expressed on tumour cells and tumour-infiltrating immune cells, blocking its interactions with both PD-1 and B7.1 receptors. By inhibiting PD-L1, Tecentriq may enable the re-activation of T cells. Tecentriq may also affect normal cells. Tecentriq has been approved for some of the most aggressive and difficult-to-treat forms of cancer, and is the first PD-(L)1 cancer immunotherapy available in both subcutaneous and intravenous formulations. From Phase III Trials to Commercialization Partnerships, MindRank's AI Drug Development Value Comes into Focus

Hong Kong, Oct 9, 2026 - As AI Drug Development Approaches Commercialization, MindRank Marks a Key Business Milestone How much value can AI create in the discovery and development of new medicines' For investors following the sector, the answer lies in tangible progress in drug development and business execution. A platform’s ability to consistently generate drug candidates, advance lead assets through clinical development and secure recognition from industry partners provides an important basis for assessing an AI drug developer’s growth potential. The recently announced commercialization partnership by MindRank AI Ltd. (MindRank, hereinafter “MindRank”) offers a new window into this question. On September 28, 2026, MindRank announced an exclusive commercialization partnership with 3SBio (01530.HK) subsidiaries Sunshine Mandi and Wansheng Pharmaceutical for MDR-001, its internally developed, AI-designed oral small-molecule GLP-1 receptor agonist. Covering mainland China, Hong Kong and Macau, the agreement carries a potential total transaction value of up to RMB1.75 billion, alongside a substantial share of sales revenue. MDR-001 is currently undergoing a pivotal Phase III clinical trial in China in adults with obesity or overweight. For MindRank, a clinical-stage AI-native drug discovery and development technology company, the partnership further connects drug development with commercialization planning. It lays the groundwork for marketing its lead asset following potential approval and establishes a clearer pathway for translating AI-driven research into commercial value. As its lead asset advances through pivotal clinical development, MindRank has begun preparing for post-approval marketing and channel coverage, extending the product’s development pathway into commercialization. Under the agreement, Sunshine Mandi and Wansheng Pharmaceutical will provide exclusive, comprehensive services and distribution across all channels following MDR-001’s launch. MindRank will retain all rights, title and interests in the product’s intellectual property and remain responsible for clinical development, regulatory submissions, manufacturing and supply. The transaction includes up to RMB150 million in payments for collaboration rights, up to RMB1.6 billion in royalty payments upon the fulfillment of agreed conditions, and a substantial share of sales revenue. From a business-model perspective, the agreement establishes a clear division of responsibilities. MindRank will continue investing in research and product development, while its partners will provide exclusive, comprehensive services and distribution across all channels within the agreed territories following regulatory approval and launch. In an industry that requires sustained investment, this collaboration could help MindRank retain its core asset rights while drawing on its partners’ commercial capabilities to support launch preparations and focus its own resources on strengthening its core capabilities. The partners’ willingness to make commercialization arrangements for an investigational drug also provides a new reference point for assessing its potential industry value. Clinical data address whether a product merits further development, while commercial partnerships indicate whether industry participants are willing to take part in its subsequent development. Together, these elements make MindRank’s growth story more tangible. A Lead Asset in Phase III Provides a Clearer Basis for Assessing AI-Driven R&D Capabilities The core asset underpinning the partnership is MDR-001, which continues to advance through development. In February 2026, MindRank announced that the first participant had been dosed in MOBILE, its Phase III clinical trial of MDR-001. According to the company, the multicenter, randomized, double-blind, placebo-controlled study plans to enroll approximately 750 participants across around 50 clinical centers in China to further evaluate the product’s long-term efficacy and safety. This means the evidence available to assess MindRank’s AI-driven R&D capabilities now extends to larger-scale, late-stage clinical research. For investors, the lead asset’s entry into Phase III sharpens the focus on several key questions: whether long-term efficacy can be further validated, whether safety and tolerability findings can be sustained, and whether the product can establish clinically meaningful competitive advantages. Previously disclosed Phase IIb findings supported further development. In the study, which enrolled 317 adults with obesity or overweight, participants in the MDR-001 dose groups achieved mean reductions in body weight from baseline of 8.2% to 10.3% after 24 weeks, compared with 2.5% in the placebo group. The rate of treatment discontinuation due to treatment-emergent adverse events was 0.8%. Common adverse events were primarily gastrointestinal and mostly mild to moderate. These findings remain subject to further validation in subsequent studies. For weight-management medicines, product value depends on a combination of weight-loss efficacy, long-term safety, tolerability and ease of use. Compared with the injectable peptide-based GLP-1 medicines that currently dominate the market, MDR-001’s oral small-molecule formulation can significantly improve medication adherence, while its large-scale manufacturing costs are substantially lower than those of peptide-based medicines, offering considerable market potential and commercial value. These attributes provide a promising direction for its future product positioning, although translating them into a competitive advantage will require continued accumulation of clinical evidence. Beyond progress in product development, R&D efficiency is another important lens through which to assess MindRank. According to the company, MDR-001 progressed from project initiation to preclinical candidate (PCC) nomination in just eight months, during which approximately 80 small molecules were synthesized. Its investigational new drug (IND) application was cleared in the United States within 19 months of project initiation and MDR-001 advanced to Phase III in China in approximately 4.5 years. In its July 2026 financing announcement, the company further disclosed that cumulative direct R&D investment in the program, from initiation through the start of Phase III in China, totaled approximately US$23 million. These timelines and investment figures provide a more concrete basis for understanding how MindRank organizes drug discovery and development. For drug developers, development timelines and funding requirements directly affect how many programs they can advance, how long they can sustain development and how they allocate resources to subsequent work. AI’s commercial significance must also be assessed within these real-world R&D constraints. As a flagship application of MindRank’s AI-driven drug discovery and development system, MDR-001 offers a traceable development pathway: from molecular design to clinical research and, subsequently, commercialization partnerships. The platform’s capabilities are beginning to be tested through tangible business outcomes. This provides capital markets with a more useful basis for understanding the company’s technology value than descriptions of algorithms or model parameters alone. Beyond the Lead Asset, Long-Term Growth Depends on Sustained Platform Output MDR-001 is the central focus of MindRank’s business progress at this stage, while the company’s long-term growth potential also depends on its ability to continue developing new drug assets. According to the MindRank, its  end-to-end AI drug development engine, MAP (Molecule Arts Platform), integrates biology, chemistry, computation, experimental validation and clinical learning into a unified drug discovery and development system. In July 2026, the company announced that it had completed a cumulative US$52 million Series B financing in stages, with proceeds earmarked for further platform development and the advancement of its clinical and preclinical pipeline. In September 2026, the company formally launched Intera (Intelligence Era), its life-sciences foundation model system, and released a technical report on InteraFold-Chem, the first model in the InteraFold series. From an operating perspective, platform development and pipeline advancement have the potential to reinforce each other. R&D programs bring real-world problems and experimental feedback to the platform, while platform iterations support the discovery and optimization of subsequent programs. Ultimately, the platform’s value must be reflected in sustained output: its ability to advance more molecules with development potential across different targets and disease areas, gradually building a business supported by multiple assets. MindRank has already expanded beyond metabolic diseases. In March 2025, the company disclosed that MRANK-106, an oral dual inhibitor of WEE1 and YES1, had received IND approval in the U.S. in oncology. This development provides another case through which to assess the applicability of its AI platform across different R&D areas, although the product’s potential remains subject to subsequent clinical validation. In earlier-stage research, the company disclosed in April 2026 that a collaborative study on drug discovery for Alzheimer’s disease, conducted by MindRank, the University of Macau and Imperial College London, had been published in Nature Biomedical Engineering. The work combined AI screening with experimental validation to identify small molecules meriting further investigation, demonstrating the company’s research capabilities and exploration at the forefront of drug discovery for complex diseases. These programs are at different stages, and their contributions to the company’s value need to be assessed separately. MDR-001 remains the main focus of current clinical development and commercialization preparations, while other pipeline programs and research support the platform’s broader applicability and long-term project portfolio. As more programs deliver verifiable progress, outside observers will be better positioned to assess whether the company can consistently replicate its R&D capabilities. Viewed through this lens, the recent commercialization partnership offers an opportunity to reassess MindRank. Its lead asset has entered pivotal clinical development, partners are participating in commercialization preparations at an early stage, financing supports continued R&D, and its presence across multiple areas preserves opportunities for future growth. These interconnected developments make the company’s development trajectory easier for capital markets to understand. MDR-001 remains in clinical development and has not yet received marketing approval. Its future value realization depends on clinical results, regulatory approval and commercial execution. Nevertheless, the business progress achieved to date provides a more concrete basis for assessing MindRank. For investors following opportunities at the intersection of AI and innovative medicines, the central question worth tracking is whether the company can progressively translate its established R&D capabilities into more verifiable drug assets and commercial outcomes.
