Darolutamide plus ADT and radiation therapy reduces risk of metastasis or death by 58% in prostate cancer patients at high risk of recurrence

Berlin, October 24, 2026 – New late-breaking data to be presented today at the European Society for Medical Oncology (ESMO) Congress 2026 will show that adding Bayer’s androgen receptor inhibitor (ARi), darolutamide, to standard-of-care androgen deprivation therapy (ADT) and locoregional radiation therapy (RT), significantly improves metastasis-free survival based on conventional imaging (MFS-CI) in prostate cancer patients with high risk of recurrence compared to placebo. Darolutamide is currently not approved for this treatment setting. These data will be presented at Presidential Symposium I at the ESMO Congress 2026 (LBA4, Saturday 24 October, 16:30 CET). At a median follow-up of 3.8 years, the interim analysis demonstrated a 58% reduction in the risk of metastasis or death with darolutamide plus ADT and RT compared with placebo plus ADT and RT (hazard ratio [HR] 0.42; 95% CI: 0.27–0.66; p=0.0001). The 4-year metastasis-free survival rate was 95% for patients receiving darolutamide compared with 88% for those receiving placebo. “Prostate cancer patients at a high risk of relapse face a considerable risk of recurrence and metastatic spread, making early, highly effective intervention vital,” said Dr. Tamim Niazi, Associate Professor of Oncology at McGill University, Radiation Oncologist at the Jewish General Hospital in Montreal, Canada. and Co-Lead Investigator of the study. “The clear improvement in metastasis-free survival seen in interim results from the DASL-HiCaP trial highlights the potential of adding darolutamide to standard androgen deprivation therapy and radiation therapy to help prevent or delay disease progression in patients who are at high risk of progression despite receiving primary curative therapy.” Secondary endpoints also showed consistent positive trends with darolutamide, with a 53% reduction in the risk of prostate-specific antigen progression free survival (PSA-PFS; HR 0.47; 95% CI: 0.33–0.67), and a 4-year PSA-PFS rate of 91% versus 83% with placebo. Prostate cancer-specific survival (PCSS) and overall survival (OS) hazard ratios were 0.58 (95% CI: 0.17–1.97; 99% vs 99% at 4 years) and 0.88 (95% CI: 0.49–1.59; 97% vs 95% at 4 years), respectively, with follow-up ongoing. The safety profile of darolutamide was consistent with its known tolerability, with adverse events being similar to the placebo group. Proportions of patients experiencing Grade ≥2 adverse events of special interest for darolutamide versus placebo were: fatigue/asthenia (14% vs 10%), hypertension (7% vs 9%), fractures (2% vs 3%), and cardiac events (3% vs 3%). Any Grade ≥3 adverse event occurred in 43% of patients in the darolutamide group versus 36% in the placebo group. "These new data highlight the potential of darolutamide to make a meaningful difference for patients earlier in the course of their prostate cancer,” said Christine Roth, Executive Vice President, Global Product Strategy and Commercialization at Bayer’s Pharmaceuticals Division. “By advancing darolutamide into earlier disease settings, we aim to reach more patients across the prostate cancer care continuum. We are proud to support ANZUP, the Sydney Clinical Trials Centre, CCTG and international partners in research as we work together with the potential to improve outcomes for men living with prostate cancer.” Follow-up of participants is continuing to provide further information on longer-term outcomes, including overall survival. About the DASL-HiCaP trial (ANZUP1801)  The DASL-HiCaP trial is an investigator-initiated, randomized, placebo-controlled, Phase III trial (NCT04136353) evaluating darolutamide added to standard-of-care ADT and locoregional radiation therapy in prostate cancer patients at high risk of recurrence. DASL-HiCaP was conducted in collaboration with the University of Sydney and ANZUP Clinical Trials Group and is the fourth Phase III trial for darolutamide. Following ARAMIS, ARASENS, and ARANOTE, which demonstrated darolutamide’s efficacy and safety in advanced prostate cancer, DASL-HiCaP represents the first Phase III study of darolutamide in early prostate cancer. A total of 1,106 participants across Australia, New Zealand, Canada, the United States, Ireland and the United Kingdom were randomized 1:1 to receive darolutamide (600 mg twice daily) or matching placebo for 96 weeks, combined with an LHRH analog for 96 weeks and RT to the prostate/prostate bed and pelvic lymph nodes. The primary endpoint was metastasis-free survival based on conventional imaging (MFS-CI). Secondary endpoints included prostate-specific antigen progression free survival (PSA-PFS), prostate cancer-specific survival (PCSS), and overall survival (OS). The protocol specified an interim analysis after 87 MFS-CI events; follow-up is ongoing to evaluate long-term overall survival. About the Sydney Clinical Trials Centre The Sydney Clinical Trials Centre is a flagship research centre of the Faculty of Medicine and Health at the University of Sydney that designs and manages clinical trials. This includes responsibility for study coordination, monitoring, data acquisition and management and statistical analysis. The Sydney Clinical Trials Centre has health economics, biostatistics, systematic reviews and biomarker teams that work with trial data and inform healthcare providers about best practice. ANZUP receives valuable infrastructure support from the Australian Government through Cancer Australia and research funding from Prostate Cancer Foundation of Australia. About Prostate Cancer Prostate cancer is the second most common cancer and the fourth most common cause of cancer death in men worldwide. Over 1.5 million men are diagnosed with prostate cancer each year and almost 420,000 die from the disease worldwide. By 2040, prostate cancer diagnoses are projected to increase to 2.9 million. Standard therapy for prostate cancer patients at high risk of recurrence combines radiation therapy with androgen deprivation therapy (ADT). However, despite this approach, a significant proportion of patients experience treatment failure, local recurrence, or distant metastases. Intensifying therapy early with an advanced androgen receptor inhibitor aims to improve long-term disease control and delay metastatic progression. About darolutamide (Nubeqa™) Darolutamide is an oral ARi with a distinct chemical structure that binds with high affinity to the androgen receptor and exhibits a strong antagonistic effect against the androgen receptor inhibiting the receptor function and growth of prostate cancer cells. Additionally, preclinical models and neuroimaging data in healthy humans support darolutamide's low potential for blood-brain barrier penetration and low potential for clinically relevant drug-drug interactions. Darolutamide, under the brand name Nubeqa™ plus androgen deprivation therapy (ADT), is already approved in more than 85 markets around the world, including the US, EU and China. It is approved for the treatment of adult patients with mHSPC without docetaxel in 75 markets and with docetaxel in more than 85 countries, and for adult patients with nmCRPC at high risk of developing metastatic disease in more than 85 countries. Darolutamide is developed jointly by Bayer and Orion Corporation, a globally operating Finnish pharmaceutical company. Darolutamide demonstrated efficacy and tolerability in pivotal Phase 3 studies, supporting its indications in nmCRPC and mHSPC with and without chemotherapy, with a favorable safety profile that offers an advancement in treatment management for physicians and patients. A robust clinical development program is underway investigating darolutamide across various stages of prostate cancer. Current ongoing trials include the Phase III ARASTEP trial evaluating the efficacy of darolutamide plus ADT compared to ADT alone in hormone-sensitive prostate cancer (HSPC), in patients with high-risk biochemical recurrence (BCR) who have no evidence of metastatic disease by conventional imaging and a positive PSMA PET/CT at baseline. France's Debt Crisis Sparks Nationwide Protests Amid Eurozone Instability

Fredericksburg, TX Friday Oct 23, 2026 @ 10:25 PM Central — Episode 1913 of the No Agenda Show, titled 'Meat Muck' and published October 18, 2026, finds host Adam Curry broadcasting from the Texas Hill Country alongside guest co-host Rob Dew, dubbed Sir Deucifer. Recorded the morning after the fifth annual Fredericksburg meetup, the episode tackles a packed slate of global unrest and domestic strangeness, from burning French city halls to Elon Musk's hopping Roadster. With John C. Dvorak's absence still shaping the program, Curry and Dew deliver the trademark media deconstruction that has defined the show for 19 years. The two-hosts spend the opening segment on the escalating protests across France, where interior minister Laurent Nunez reported crowds swelling to 800,000 and Marine Le Pen's poll numbers continue climbing against a backdrop of debt projected near 120% of GDP. Threads explored include: How protesters are coordinating via Bluetooth mesh apps like BitChat after internet shutdowns AfD leader Alice Weidel's call to end German financing of 'an already lost war' in Ukraine Queen Ursula von der Leyen's new 70 billion euro Ukraine funding gap for 2027 The Nord Stream sabotage trial of Ukrainian suspect Sergei Kuznetsov in Hamburg Curry questions why the International Criminal Court has stayed silent on the Nord Stream prosecution, while Dew notes the political convenience of keeping the accused agent from speaking. The hosts replay a Christopher Hitchens clip framing the broader civilizational stakes: They are not just joking. They really mean to abolish everything you care about, and they want to take away everything you love and to destroy everything you have and to replace it with a Stone Age ideology derived from the desert of Palestine in a very bad time in human history. The segment pivots to a Tom Luongo thesis that European elites are deliberately bankrupting the bloc to roll out the digital euro. Elsewhere, the episode unpacks an AI-driven study flagging roughly 261,000 potentially fabricated cancer research papers, with 36% traced to Chinese institutions and 20% to Iran. Dew brings a deep dive on Turin, Italy, its black magic triangle, ley lines to London and San Francisco, and the 1879 Lucifer statue atop the Frejus tunnel monument. Curry breaks down CoreWeave's 2032 bonds trading at distressed 15% yields, warning the AI buildout could trigger a broader bond market crack. Other threads: Tesla's thruster-equipped Roadster demo, Press Secretary Katie Zachariah's five-day White House tenure, Jalen Hurts' blood clot diagnosis, and viral video evidence that Arby's roast beef now arrives as a bagged paste. About No Agenda Show No Agenda is a long-running, listener-supported podcast hosted by Adam Curry and John C. Dvorak that takes a skeptical, independent look at mainstream media, politics, culture, and the forces shaping the daily news cycle. Known for its sharp commentary, humor, and media deconstruction approach, the show offers an irreverent alternative to conventional news analysis. Episode 1913 is available now wherever podcasts are heard.

ZenaTech Unveils its Proprietary AI Software Suite Designed to Accelerate Land Survey Delivery for Drone as a Service Commercial, Municipal and Infrastructure Clients

Vancouver, BC, Oct 24, 2026 - ZenaTech, Inc. (Nasdaq: ZENA)(FSE: 49Q)(BMV: ZENA)("ZenaTech" or the "Company"), a technology solution provider specializing in AI (Artificial Intelligence) drone, Drone as a Service (DaaS), enterprise SaaS, and Quantum Computing solutions, today unveiled its proprietary artificial intelligence software suite and planned integration across its DaaS land survey operations. By pairing ZenaDrone high-density LiDAR (Light Detection and Ranging) data acquisition with automated feature extraction and point cloud classification, ZenaTech plans to accelerate survey-grade data delivery time from days to as little as minutes across commercial land surveying, municipal and infrastructure projects for DaaS clients across North America and around the world. The combined platform is currently in the development and testing stage. ZenaTech, Inc. is a B2i Digital Featured Company. See the company’s in-depth profile at: https://b2idigital.com/zenatech-1   “Land survey delivery has traditionally been limited by manual post-processing, which caps how many projects a survey team can complete,” said Shaun Passley, Ph.D., CEO of ZenaTech. “By integrating our AI software directly into ZenaDrone DaaS operations, we aimto automate the heavily manual work of point cloud classification and CAD linework generation, so the same crews and drones can deliver turn-key, actionable intelligence to clients faster and with consistent quality. As we apply our AI suite across our AI tools growing network of DaaS subsidiaries, we believe it can improve operating efficiency and support the scalable, recurring revenue model at the center of our DaaS strategy.” The combined platform is designed to bridge hardware data collection with AI-driven point cloud processing. A ZenaDrone land survey drone equipped with high-density LiDAR captures a site as a point cloud, a dataset that can contain billions of individual 3D measurement points. ZenaAI is then intended to run automated geospatial workflows on that data, classifying each point by what it represents, converting the classified data into terrain models and vector drawings, removing noise, tagging infrastructure assets and exporting the results in formats that survey and engineering teams can use directly. Its key planned capabilities include: Automated Point Cloud Classification: Designed to use deep learning neural networks to rapidly parse billions of 3D LiDAR points, separating ground returns from dense canopy, buildings, utility lines and surface vegetation to generate true Digital Terrain Models (DTM) Automated Feature Extraction and Linework: Computer vision algorithms are intended to detect and vectorize critical planimetric features, including curb lines, pavement edges, retaining walls and structures, and export them directly into CAD and BIM (Building Information Modeling) formats Rapid DaaS Turnaround: By replacing manual point-picking and heuristic scripting, the AI software suite is being designed to reduce post-processing workflows by as much as 80% and shorten survey turnaround time, allowing DaaS survey teams to deliver fully classified datasets within hours of flight completion Smart Noise Filtering and Asset Tagging: Automated artifact suppression is intended to clean airborne noise, multipath errors and scan overlap, while semantic tagging logs discrete infrastructure assets, such as manholes, hydrants and utility poles, with millimeter-level precision Point cloud classification and linework have traditionally required skilled technicians to work through by hand, which can take days and limits how many projects a survey team can complete. ZenaTech believes that automating these steps could raise surveying productivity, allowing the same drones and crews to deliver more projects faster and potentially improving margins over time. As proprietary software, the AI suite is also intended to add a scalable SaaS layer to the Company’s acquisition-led DaaS strategy, applying a consistent processing workflow across the surveying businesses it acquires and supporting its recurring revenue model for commercial, municipal and infrastructure clients that rely on timely data for planning, design and construction.   The AI-powered LiDAR processing is currently in development with testing across DaaS land surveying, infrastructure monitoring and utility inspection projects expected next year. The Company will provide updates on launch and rollout as milestones are achieved. ZenaTech’s Drone as a Service (DaaS) division provides on-demand turnkey drone piloting, data capture, and data processing services to commercial, government, and infrastructure customers, allowing them to access aerial data and automation without owning or operating their own drone fleets. The division is being built through an acquisition-led strategy that combines local low-tech service providers into a global network, creating a scalable platform for delivering recurring-revenue aerial and data services such as surveying, inspections, power washing, and construction progress monitoring. About ZenaTech ZenaTech, Inc. (Nasdaq: ZENA) (FSE: 49Q) (BMV: ZENA) is a technology company which specializes in AI autonomy drone platforms to transform commercial, government, and defense sectors. Roche presents interim phase III data showing sefaxersen significantly reduced proteinuria in people with IgA nephropathy (IgAN)

Sefaxersen achieved a statistically significant 43.1% reduction in proteinuria, a key indicator of kidney damage, relative to placebo at 37 weeks with consistent treatment effects across all prespecified patient subgroups1 As the first antisense oligonucleotide (ASO) evaluated in IgAN, sefaxersen inhibits complement factor B production and has the potential to slow IgAN disease progression by reducing kidney inflammation2 Investigational sefaxersen is intended to be a once-monthly subcutaneous injection and designed to enable convenient self-administration at home3 Despite recent advances, many people with IgAN remain at risk of disease progression and kidney failure, highlighting the need for additional targeted treatment options for those who do not respond to initial therapy4,5 Basel, 23 October 2026 - Roche (SIX: RO, ROP; OTCQX: RHHBY) announced today late-breaking oral presentation data from the ongoing phase III IMAgINATION study in adults with primary IgA nephropathy (IgAN) receiving optimised standard of care, demonstrating that investigational sefaxersen achieved a statistically significant and clinically meaningful 43.1% reduction in proteinuria (95% confidence interval [CI]: 32.5 - 52.0, P<0.0001), relative to placebo at 37 weeks, as measured by 24-hour urine protein-to-creatinine ratio (UPCR).1 Proteinuria is a key indicator of kidney damage and a reduction in UPCR is strongly associated with the preservation of long-term kidney function.6,7 Results presented at the American Society of Nephrology (ASN) Kidney Week 2026 also showed that, among patients with baseline haematuria (blood in urine), a greater proportion of patients achieved haematuria resolution at Week 37 with sefaxersen versus placebo.1 “These phase III results demonstrate the potential for sefaxersen to meaningfully reduce protein in the urine, a key indicator of kidney damage, in people with IgA nephropathy,” said Levi Garraway, MD, PhD, Roche's Chief Medical Officer and Head of Global Product Development. “By inhibiting complement factor B in the liver, sefaxersen may offer a novel approach to slow disease progression in IgAN, potentially helping patients delay or avoid long-term dialysis or kidney transplantation.” The treatment effect in all prespecified subgroups was consistent with the clinically meaningful reductions in proteinuria shown in the primary endpoint results.1 Sefaxersen was well tolerated with a safety profile consistent with the previously reported phase II study, exhibiting no new safety signals.1,2 Sefaxersen is an investigational once-monthly subcutaneous injection designed to enable convenient self-administration for people with IgAN.3 The IMAgINATION study will continue as a blinded study to evaluate the change in kidney function over two years, as measured by estimated glomerular filtration rate (eGFR) at week 105. Data will be shared with health authorities, with the goal of bringing this treatment to patients as soon as possible. Typically diagnosed before the age of 40, IgAN affects at least 25 adults per million worldwide each year.8,9 IgAN, also known as Berger’s disease, is a chronic and progressive autoimmune disease that leads to end-stage kidney disease in up to 50% of patients within 20 years of diagnosis.5,10 It is the most common form of primary glomerulonephritis (inflammation of the tiny filters, glomeruli, in the kidneys) and is a major cause of chronic kidney disease as well as kidney failure.8 About sefaxersen Investigational sefaxersen is the first highly specific liver-directed antisense oligonucleotide (ASO) inhibitor of factor B production being evaluated in IgA nephropathy (IgAN).2 Serum complement factor B levels are elevated in people with IgAN. When we reduce these levels, it in turn reduces the level of protein in urine, which is a key indicator of improved kidney function.11,12 Sefaxersen is designed to provide sustained control of the alternative complement pathway, an important pathway in IgAN, enabling once-monthly subcutaneous dosing and is intended for self-administration in people with IgAN.3 In phase I and II trials, sefaxersen was shown to be generally well tolerated in healthy volunteers and people with IgAN who were at high risk of progression, and reduced proteinuria and plasma complement factor B.2,13 Roche licensed sefaxersen from Ionis Pharmaceuticals for the treatment of complement mediated diseases. About the IMAgINATION study IMAgINATION [NCT05797610] is a phase III, multicentre, randomised, double-blind, placebo-controlled study to evaluate the efficacy and safety of subcutaneous sefaxersen, an investigational antisense inhibitor of complement factor B, in patients with primary IgA nephropathy at high risk of progression.1,14 The study enrolled 459 people, who are randomised 1:1 to receive sefaxersen or placebo for 105 weeks.1,14 Participants may switch to open-label treatment after week 105 at the investigator's discretion or after the common-close timepoint, whichever occurs first.1,14 The primary endpoint of the study is the change from baseline in the urine protein-to-creatinine ratio at 37 weeks.1,14 About primary immunoglobulin A nephropathy (IgAN) IgAN, also known as Berger’s disease, is a serious, chronic and progressive autoimmune kidney disease affecting at least 25 adults per million worldwide each year and is typically diagnosed before the age of 40 years.8-10 In IgAN, immune complexes deposit in the kidneys, which activates the complement system’s alternative pathway, leading to inflammation and subsequent kidney damage.15 It is the most common form of primary glomerulonephritis (inflammation of kidney glomeruli) and remains a leading cause of kidney failure, with up to 50% of patients experiencing end-stage kidney disease within 20 years of diagnosis.5,8 Many current therapies aim to reduce protein levels in the urine and control blood pressure to slow progression of the disease.16 Updated KDIGO 2025 guidelines* highlight the need to address both the underlying immune-mediated causes of IgAN and the downstream consequences of kidney damage.4 While new therapies have expanded treatment options, important unmet needs remain, including improved long-term preservation of kidney function and more targeted approaches that address key drivers of disease progression.4,16 About Roche in Immunology For over two decades, Roche has advanced immune system research, creating groundbreaking targeted treatments that have transformed care for people with blood disorders, neurological conditions and autoimmune diseases. With a development pipeline exploring more than 20 medical approaches, we are targeting widespread, complex conditions like chronic lung disease (COPD) and inflammatory bowel disease (IBD), alongside lupus and autoimmune kidney diseases. Our ultimate goal is to lessen the burden of chronic immune disorders on patients, healthcare systems, and society. Global Business Series

Friday 23 October, 2026 The Global Business Series is a collaboration between leading academics at Tsinghua University, China (12th in World University Rankings 2026) and the London School of Economics. In the series. Six distinguished academics tackle four different aspects of doing business internationally – from family businesses to digital platforms to law and marketing. Critical Decisions The authors examine critical issues around succession, continuity and inheritance – issues which every family business will eventually face. Through detailed research the authors provide original insights and lessons. Anyone playing a key role in a family-owned business will benefit from reading this insightful book. The authors: “The reason why the development of family businesses is bumpy and the succession issues are complicated and difficult is that family businesses… involve not only the logic of business and economic operations, but also multiple social and cultural relationships.” Platform Governance In a world where our existence is largely platform-based – from shopping on Amazon to socializing on Facebook – understanding the mechanisms governing these digital habitats is paramount. This authoritative guide takes you from the origins of these platforms to the strategic shift required to move from a traditional product strategy to a winning platform strategy.  It offers vital insights and guidance for understanding and succeeding in today’s digital economy, helping readers navigate the complexities of an increasingly platform-based economy. Think Big Using legal thinking, Han Liu looks at how to analyse situations, balance competing interests and make good decisions – both in business and more broadly in life and everyday situations. This book will empower readers to think critically and strategically. It will help shift mindsets from simply following rules to understanding the deep, strategic logic behind them. It's a journey that will sharpen minds, helping to make better, more informed choices. “If you’re worried about legal jargon, don’t be. This book explains legal thinking in everyday language.” – Han Liu, from the foreword Wellbeing Marketing Examines transformative philosophy to benefit customers and employees. When maximising profit has often become a demon justifying deceptive tactics, exploited workers and unhappy customers, Wellbeing Marketing emerges as a new way of thinking. Through fascinating case studies of global giants like Disneyland and Walmart, alongside unique enterprises like Nakagawa Masashichi Heirloom Store, Fei Li demonstrates that profit and happiness are not mutually exclusive; rather, when an organization aligns its mission with the pursuit of wellbeing, it achieves a sustainable "unity of knowledge and action" that benefits society. About the series authors: Dr Hao Gao is Director of the Research Centre for Global Family Business at Tsinghua University, China. Dr Victor Zheng is Director of the Centre for Social and Political Development Studies at the Chinese University of Hong Kong (CUHK).
